eflornithine; difluoromethylornithine (Vaniga, Ornidyl)
Tradename: Vaniga (topical)
Indications:
1) Trypanosoma brucei-gambiense
2) variable efficacy against Trypanosoma brucei-rhodesiense
3) topical: reduce growth of unwanted facial hair
4) hirsuitism [4,5]
Dosage:
1) systemic
- 400 mg/kg/day IV in 4 divided QID for 2 weeks
- followed by 300 mg/kg/day PO for 3-4 weeks
- 90% cure rate for stage II disease (abnormal CSF)
2) topical
Pharmacokinetics:
-> topical (13.9%)
a) may take up to 8 weeks for effects
b) effective in 1/2 of women who try it
Adverse effects:
1) diarrhea
2) anemia
3) thrombocytopenia
4) seizures
5) hearing loss
Mechanism of action:
-> inhibits enzyme to slow hair growth
