Function: - receptor for endothelin-1 - mediates its action by association with G proteins that activate a phosphatidylinositol-Ca+2 second messenger system - The rank order of binding affinities for ET-A is: ET1 > ET2 >> ET3 - interacts with HDAC7 & HTATIP Structure: belongs to the G-protein coupled receptor 1 family Compartment: cell membrane Alternative splicing: named isoforms=4 Expression: - isoforms 1, 3 4 are expressed in a variety of tissues: aorta, cerebellum > lung, atrium, cerebral cortex > placenta, kidney, adrenal gland, duodenum, colon, ventricle & liver; no expression in umbilical vein endothelial cells - within the placenta, isoforms 1,2,3 & 4 are expressed in the villi & stem villi vessels
