Function: - receptor for TNFSF12/APO3L/TWEAK - interacts directly with the adapter TRADD - mediates activation of NF-kappa-B & induces apoptosis - may play a role in regulating lymphocyte homeostasis - interacts strongly via the death domains with TNFRSF1 & TRADD to activate at least 2 distinct signaling cascades, apoptosis & NF-kappa-B signaling. - interacts with BAG4 Structure: - homodimer - glycosylated (probable) - contains 1 death domain - contains 4 TNFR-Cys repeats Compartment: - isoforms 1,2,9,11: cell membrane - isoforms 3,4,5,6,7,8,10,12: secreted Alternative splicing: named isoforms=12 Expression: - abundantly expressed in thymocytes & lymphocytes - detected in lymphocyte-rich tissues incuding: thymus, colon, intestine, & spleen - also found in the prostate Note: - some isoforms may be produced at very low levels due to a premature stop codon in the mRNA, leading to nonsense-mediated mRNA decay
