CDK2/cyclin E complex

Function: - the cyclin E/CDK2 complex controls entry into S phase - CDK2 is expressed constitutively throughout the cell cycle - association of CDK2 with cyclin A or cyclin E with subsequent dephosphorylation of Tyr15 & Thr14 on CDK2 by cdc25 phosphatase & an independently regulated phosphorylation of Thr160 activates CDK2 - CDK2/cyclin-E is maximally activated a few hours before onset of DNA synthesis - CDK2 activity is required for DNA synthesis & is responsible for phosphorylation of Rb protein & modulation of DNA-bound replication protein A
