Function: 1) component of ESCRT3 in the multivesicular body pathway 2) in the ESCRT3 complex, it interacts directly with CHMP4A, CHMP4B, CHMP6 3) self-associates 4) interacts with PDCD6IP of ESCRT1 complex 5) interacts with CHMP2A 6) interacts with VPS4A Structure: - the acidic C-terminus & the basic N-termminus are thought to render the protein in a closed, soluble & inactive conformation through an autoinhibitory intramolecular interaction - the open & active conformation, which enables membrane binding & oligomerization, is facilitated by interaction with other ESCRT components (putative) - belongs to the SNF7 family Compartment: cytoplasm, late endosome membrane Expression: expressed in heart, spleen, kidney Pathology: - the HIV-1 virus takes advantage of ESCRT3 for budding & exocytosis of viral proteins, via the association of CHMP4 proteins with PDCD6IP, a protein directly recruited by HIV-1 p6 protein that functions at sites of viral Gag assembly & budding - overexpression strongly inhibits HIV-1 release
