Function: - component of ESCRT3 in the multivesicular body pathway - within ESCRT3, serves as an acceptor for ESCRT1 complex - interacts with CHMP2A, CHMP4B, CHMP4C, CHMP6 within ESCRT3 - interacts with ESCRT1 component PDCD6IP - self-associates - overexpression leads to the assembly of filaments that curve & associate to create circular rings that can promote outward budding - interacts with VPS4A Structure: - the acidic C-terminus & the basic N-termminus are thought to render the protein in a closed, soluble & inactive conformation through an autoinhibitory intramolecular interaction - the open & active conformation, which enables membrane binding & oligomerization, is facilitated by interaction with other ESCRT components (putative) - belongs to the SNF7 family Compartment: - cytoplasmic, membrane-associated - localizes to large vesicle-like structures - localizes to the midbody of dividing cells - localized in two distinct rings on either side of the midbody Expression: - widely expressed - expressed in heart, kidney, liver, skeletal muscle > brain, placenta, lung, pancreas Pathology: - role in HIV-1 virus release via its interaction with PDCD6IP
