CD43; leukosialin; leukocyte sialoglycoprotein; sialophorin; galactoglycoprotein; GALGP (SPN)
Function:
- one of the major glycoproteins of thymocytes & T-cells
- role in the physicochemical properties of the T-cell surface
- role in lectin binding, presents carbohydrate ligands to selectins
- counter receptor for CD169 (putative)
- during T-cell activation is actively removed from the T-cell to antigen-presenting cell contact site, thus a negative regulatory role in adaptive immune response (putative)
- interacts with HIPK2 via the cytoplasmic domain
Structure:
- has an extended rodlike structure that could protrude above the glycocalyx of the cell & allow multiple glycan chains to be accessible for binding
- glycosylated, has a high content of sialic acid & O-linked carbohydrate structures
Compartment: membrane
Expression:
- T cells
- activated B cells
- NK cells
- granulocytes
- monocytes
- megakaryocytes
- brain
Pathology:
1) expressed in:
a) T cell lymphomas ~80%
b) coexpressed with CD20 in small lymphocytic lymphoma
c) granulocytic sarcomas
d) AML
e) ALL (most)
f) plasmacytomas
g) mast cell disease
2) defective expression seen in Wiskott-Aldrich syndrome
