Function:
1) innate immune response to microbial agents
2) recognition of dsRNA of viral origin (putative)
- binding of ds-RNA is required for the interaction with SRC
3) acts via MyD88 & TRAF6
a) NF-kappa-B activation
b) cytokine secretion
c) inflammatory response
4) may attenuate allergic response [3]
- stimulation decreases allergen-induced BAL eosinophils
5) binds MyD88 via their respective TIR domains
6) interacts with TICAM1
Structure:
- homodimer formation is triggered by ligand binding & is required for TLR3 signaling
- heavily N-glycosylated, except on that part of the surface of the ectodomain that is involved in ligand binding
- belongs to the Toll-like receptor family
- contains 22 LRR repeats (leucine-rich repeats)
- contains 1 TIR domain
Compartment:
- endoplasmic reticulum membrane
- endosome membrane
Expression:
- expressed at high level in placenta, pancreas > CD11c+ immature dendritic cells.
- only expressed in dendritic cells, NOT in other leukocytes
