Function: - transcriptional repressor required for germinal center formation & antibody affinity maturation - phosphorylated by MAPK1 in response to antigen receptor activation - Ser phosphorylation induces its degradation by ubiquitin/proteasome pathway - interacts with ZBTB7 & BCL6B (putative) - interacts with the catalytic domain of HDAC9 Structure: - contains 1 BTB/POZ domain - contains 6 C2H2-type Zn+2 fingers Compartment: nucleus (putative) Expression: - expressed in germinal center T & B cells & in primary immature dendritic cells - down-regulated during maturation of dendritic cells by selective stimuli such as LPS, CD40L & zymosan - CD40 activation & antigen stimulation downregulate bcl-6 Pathology: - role in lymphomagenesis (putative) - chromosomal translocation t(3;14)(q27;q32) involving BCL6 with immunoglobulin gene regions may be a cause of B-cell non-Hodgkin lymphoma - chromosomal translocation t(3;11)(q27;q23) involving BCL6 with POU2AF1/OBF1 may be a cause of a form of B-cell leukemia - chromosomal translocation t(3;4)(q27;p11) involving BCL6 with ARHH/RhoH may be a cause of non-Hodgkin lymphoma Laboratory: - BCL6 Ag in tissue
