BRCA1-associated RING domain protein 1 (BARD1)
Function:
- binds to BRCA1 & may be involved in mediating tumor suppression by BRCA1
- interacts with 50 kD subunit of cleavage stimulation factor (CSTF1) & inhibits polyadenylation
- interacts with RNA polymerase 2
- inhibits pre-mRNA 3' cleavage in vitro
- putative role in the cellular response to DNA damage
- processed during apoptosis
- the homodimer is more susceptible to proteolytic cleavage than the BARD1/BRCA1 heterodimer
- preferentially forms heterodimers with BRCA1
- can also form heterodimers with CSTF-50
Structure:
- homo- & heterodimer
- contains 3 ANK repeats
- contains 2 BRCT domains
- contains 1 RING-type Zn+2 finger
Compartment:
- nucleus
- during S phase of the cell cycle, colocalizes with BRCA1 into discrete subnuclear foci
- can translocate to the cytoplasm
Pathology:
- defects in BARD1 gene are found in primary breast cancer, ovarian cancer & uterine cancer
