Bcl-2 homologous antagonist/killer; apoptosis regulator BAK; Bcl-2-like protein 7; Bcl2-L-7 (BAK1, BAK, BCL2L7, CDN1)

Function: - promotes cell death & counteracts protection from apoptosis by bcl-2 - inhibits bcl-2 through formation of heterodimers - counteracts protection from apoptosis by adenovirus homolog E1B 19k protein - low micromolar levels of Zn+2 inhibit the promotion of apoptosis - interacts with BCL2A1 (putative) - forms heterodimers with BCL2, E1B 19k protein, & BCL2L1 isoform Bcl-XL Structure: - homodimer; formation of the homodimer is Zn+2-dependent - binding of bcl2 to Bak1 occurs via BH1 domain & BH2 domain - Bak1 also forms homodimers - intact BH3 motif is required by BIK, BID, BAK, BAD & BAX for their pro-apoptotic activity & for their interaction with anti-apoptotic members of the Bcl-2 family - belongs to the Bcl-2 family Compartment: mitochondrial outer membrane Expression: - expressed in wide variety of cell types - highest levels in the heart & skeletal muscle
