Function: - associates with HDL, & to a lesser extent VLDL & chylomicrons - thought to be important determinant of plasma triglycerides - stimulates of apo-CII lipoprotein lipase - inhibits of the hepatic VLDL-triglyceride production - activates poorly lecithin:cholesterol acyltransferase (LCAT) - phosphorylation sites are present in plasma - interacts with GPIHBP1 Structure: belongs to the apolipoprotein A1/A4/E family Compartment: secreted Expression: - expressed by liver & secreted into plasma - up-regulated by PPARA agonists - induced in early phase of liver regeneration Polymorphism: - 3 common alleles are known: allele APOA5*1, APOA5*2 & APOA5*3. Pathology: - defects in APOA5 are a cause of - susceptibility to familial hypertriglyceridemia - hyperlipoproteinemia type 5 Note: - uncertain whether Met-1 or Met-4 is the initiator
