Function: 1) role in signal transduction from Ras activation to actin cytoskeletal remodelling 2) suppresses insulin-induced promoter activities through AP1b& SRE 3) mediates Rap1-induced adhesion 4) interacts, through the N-terminal Pro-rich region, with WW domain of APBB1 5) interacts with RAP1A, PFN1, VASP & ENAH Structure: - belongs to the MRL family - contains 2 Pro-rich regions required for suppression of AP1 transcription activity - contains 1 PH domain - contains 1 Ras-associating domain Compartment: - cell membrane - colocalizes with ENA/VASP proteins at lamellipodia tips & focal adhesions, & F-actin at the leading edge - at the membrane surface, associates, via the PH domain, preferentially with the inositol phosphates, PtdIns-5-Phosphate & PtdIns-3-Phosphate - necessary for efficient interaction of RA domain with Ras-GTPases Expression: - widely expressed - high expression in thymus, spleen, lymph node, bone marrow & peripheral leukocytes - induced by all-trans-retinoic acid
