Function:
- potent mediator of new blood vessel formation
- may function as a tRNA-specific ribonuclease that abolishes protein synthesis by specifically hydrolyzing cellular tRNAs
- binds to actin on the surface of endothelial cells
- once bound, angiogenin is endocytosed & translocated to the nucleus
- angiogenin induces vascularization of normal & malignant tissues
- angiogenic activity is regulated by interaction with RNH1 in vivo
- interacts with & forms a tight 1:1 complex with RNH1
- dimerization of ANG/RNH1 heterodimer may occur
Structure:
- belongs to the pancreatic ribonuclease family
Compartment: secreted
Expression:
- mRNA expressed in wide spectrum of cells
- expressed predominantly in the liver
- also detected in endothelial cells & spinal cord neurons
- low level expression in the developing fetus,
- increased expression in the neonate
- maximal expression in the adult
Pathology:
- defects in ANG are the cause of susceptibility to familial amyotrophic lateral sclerosis type 9
Laboratory:
- ANG gene mutation
Note:
- uncertain whether Met-1 or Met-3 is the initiator
