The concept of signal transduction or signalling of extracellular signalling through 2nd messengers originated with the discovery of the role of cAMP Function: - the adenyl cyclase/cAMP-dependent protein kinase pathway mediates hormonal control of many cellular cascades including a) glycogenolysis b) lipolysis c) catecholamine synthesis - adenyl cyclases are activated through: a) Gs-alpha generated by serpentine receptor-mediated Gs protein dissociation b) protein kinase C c) Ca+2/Calmodulin d) G-beta/gamma - nearly all of the 8 cloned adenyl cyclases are regulated via interactions with the PLC pathway [2] - adenyl cyclases appear to be associated with Ca+2 channels - Ca+2 inhibition of adenyl cyclases is predicted to result in oscillations of cAMP levels - affinity for Mg+2 increases 20-fold upon activation by Gs-alpha-GTP
