Family of cell surface proteins with both potential adhesion domain & protease domain.
Function:
- cell adhesion
- sperm-egg fusion (ADAM1 & ADAM2)
- myoblast fusion (ADAM12)
- ectodomain shedding of cell surface proteins (ADAM9, ADAM10, ADAM17)
- activation of Notch (ADAM17)
- alpha-secretase activity in Alzheimer's disease (ADAM10, ADAM17)
- axon guidance
Structure:
- family of cell surface proteins with both potential adhesion domain & protease domain
- cysteine rich region & EGF domain thought to mediate cell fusion or interaction with other proteins such as chaperone proteins
- ~1/2 of ADAM proteins have metalloproteinase domain that contains a catalytic consensus sequence
Compartment: plasma membrane
Expression:
- ADAM9, ADAM10, ADAM12, ADAM15, ADAM17 & ADAM19 show widespread expression
- ADAM8 & ADAM28 are predominantly hematopoietic.
- 17 ADAMs are found in CNS.
- ADAM22 & ADAM23 are predominantly found in brain
- IL1 induces transcription of some ADAMs
