Function: - cation channel with high affinity for Na+ - gated by extracellular protons - inhibited by the diuretic amiloride - also permeable for Li+ & K+ - generates a biphasic current with a fast inactivating & a slow sustained phase - heteromeric channel assembly seems to modulate activity - homotetramer or heterotetramer with other ASIC proteins (probable) - interacts with STOM (putative) - interacts with PRKCABP & ACCN3 Structure: - belongs to the amiloride-sensitive Na+ channel family Compartment: - cell membrane; multi-pass membrane protein - localized at the plasma membrane of neurons, in the soma & punctated peripheral processes (putative) Alternative splicing: named isoforms=2 Expression: - brain & spinal cord - isoform 1 is also detected in testis, liver, colon & ovary Notes: - inhibited by anti-inflammatory agents including salicylic acid (putative) - regulated by Zn+2
