ATP-binding cassette sub-family B member 5 (P-glycoprotein ABCB5, ABCB5 P-gp, ABCB5, MDR/TAP)
Function:
1) plasma membrane transporter
2) efflux xenobiotics from cells (rhodamine dye, doxorubicin)
Structure:
- belongs to the ABC transporter family, MDR subfamily
- contains 1 ABC transmembrane type-1 domain
- contains 2 ABC transporter domains
Compartment:
- cell membrane
Alternative splicing: named isoforms=2
Expression:
- expressed by CD133-expressing progenitor cells among epidermal melanocytes (at protein level)
- widely expressed with specific expression in pigment cells
Pathology:
- expressed in several malignant tissues
- role in resistance to doxorubicin of a subset of malignant melanomas
- depletion of ABCB5 by RNAi increases sensitivity to several drugs of a subset of melanoma cells
